Epitalon
Pineal-derived tetrapeptide studied for telomerase activation, melatonin rhythm and longevity — intriguing data, mostly from one research school.
Overview
Epitalon is a four–amino-acid peptide developed by Vladimir Khavinson's group in St. Petersburg as a synthetic version of epithalamin, an extract of the pineal gland. In cell studies it activated telomerase and lengthened telomeres in human somatic cells. In rodents it extended median lifespan and reduced spontaneous tumors. In small human studies of older adults, it was associated with restored nighttime melatonin secretion, improved sleep and, in long-term cohorts, lower mortality. The research comes almost entirely from one school and has not been independently replicated in randomized trials. It is given in short courses rather than continuously, and its safety record in available literature is very clean. FDA advisors recently supported compounding eligibility, but rulemaking is still pending.
- Classification
- Synthetic tetrapeptide (Ala-Glu-Asp-Gly) modeled on pineal extract epithalamin
- Route
- Subcutaneous; intramuscular or intranasal
- Half-life
- Short (minutes); effects persist via gene expression
Mechanism of action
- Cellular uptakeShort peptide enters cells and reaches the nucleus.
- Chromatin interactionBinds DNA/histone regions, altering gene accessibility (proposed).
- Telomerase activationIncreases hTERT expression and telomerase activity in somatic cells.
- Pineal melatonin restorationNormalizes nighttime melatonin secretion in older subjects.
- Circadian and antioxidant effectsRestores rhythm and boosts antioxidant enzyme activity.
- Improved sleep and melatonin rhythm
- Telomere maintenance
- Possible lifespan extension (animal)
Researched for
- Biological aging and longevity
- Age-related melatonin decline and insomnia
- Telomere attrition
- Retinal degeneration
- Tumor prevention (animal)
Research notes
- Khavinson et al. (Bull Exp Biol Med, 2003): telomerase induction and telomere elongation in cultured human fibroblasts.
- Anisimov et al. (Mech Ageing Dev, 2003): extended median lifespan and reduced tumor incidence in aged mice.
- Korkushko et al. (2011): long-term elderly cohorts receiving pineal peptides showed improved melatonin rhythm and lower mortality — observational, single research group.
- No independent Western RCTs; the 2026 PCAC review considered insomnia as the use case.
Reported side effects
- Injection-site reactions
- Transient drowsiness or vivid dreams
- No significant adverse effects documented in available literature
Contraindications & cautions
- Active cancer (theoretical concern with telomerase activation)
- Pregnancy and breastfeeding
- Known hypersensitivity
Lab monitoring
- CBC, CMP
- Lipid panel, fasting glucose, HbA1c
- Vitamin D, hsCRP
- Melatonin level if available
- Sleep quality score (PSQI); telomere length optional
