Leuphasyl (Pentapeptide-18)
Topical enkephalin-mimicking peptide that may soften expression lines modestly — a gentle, much weaker alternative to Botox.
Overview
Leuphasyl is a five–amino-acid peptide modeled on leu-enkephalin, one of the body's natural opioid peptides. In laboratory studies it reduces the excitability of nerve cells that signal facial muscles to contract, lowering acetylcholine release at the nerve–muscle junction. In skincare it is marketed as a topical 'Botox alternative' for forehead lines and crow's feet, often combined with argireline (acetyl hexapeptide-8), which acts on a different step of the same process. Manufacturer studies report modest reductions in wrinkle depth after about a month of twice-daily use. Effects are subtle compared with injected neuromodulators, and how much reaches the muscle through intact skin is uncertain. It is very well tolerated.
- Classification
- Synthetic cosmetic pentapeptide (enkephalin analog)
- Route
- Topical
- Half-life
- Not well established
Mechanism of action
- Enkephalin receptor bindingBinds opioid-like receptors on peripheral nerve endings.
- Neuronal hyperpolarizationReduces nerve excitability and calcium channel opening.
- Reduced acetylcholine releaseLess neurotransmitter released at the neuromuscular junction.
- Muscle contraction dampenedFacial expression muscles contract less forcefully.
- Softer expression lines
- Synergy with argireline
Researched for
- Dynamic expression lines
- Forehead and crow's-feet wrinkles
- Topical neuromodulator alternative
Research notes
- Manufacturer in vitro data show reduced neuronal acetylcholine release and muscle contraction.
- Small consumer studies report wrinkle-depth reductions over 28 days, greater when combined with argireline.
- No independent, peer-reviewed randomized trials; skin penetration to muscle is unproven.
- Effects are mild and reverse when use stops; not comparable to botulinum toxin.
Reported side effects
- Mild skin irritation or redness
- Rare allergic reaction
- No systemic effects expected topically
Contraindications & cautions
- Active dermatitis or open wounds at application site
- Known hypersensitivity to formulation
Lab monitoring
- No bloodwork required for topical use
- Baseline standardized facial photography
