AOD-9604 + Hyaluronic Acid
Joint injection pairing hyaluronic acid's lubrication with AOD-9604's proposed cartilage-protective signaling; supported mainly by animal data.
Overview
This combination is a joint injection that pairs two agents. Hyaluronic acid (HA) is a natural component of joint fluid; injected HA restores lubrication and cushioning and is an established, if modest, treatment for knee osteoarthritis. AOD-9604, a growth hormone fragment, is added because animal studies suggest it supports cartilage cells and reduces degenerative changes. In a rabbit osteoarthritis model, the combination improved cartilage and mobility more than HA alone. No human trials have tested the combination. It must be given by a clinician using sterile technique, ideally with imaging guidance, and is best used within a physical-therapy-based rehabilitation plan.
- Classification
- Compounded combination: AOD-9604 peptide plus hyaluronic acid (glycosaminoglycan viscosupplement)
- Route
- Intra-articular (clinician-administered)
- Half-life
- AOD-9604 ~30 minutes systemically; HA persists in the joint for days to weeks
Mechanism of action
- Intra-articular deliveryClinician injects both agents directly into the affected joint.
- HA restores viscosityHyaluronan improves synovial lubrication and shock absorption.
- AOD signals chondrocytesProposed support of cartilage cells and matrix production (animal data).
- Catabolic signaling reducedLower inflammatory mediators and matrix-degrading enzyme activity reported.
- Cartilage preservedSlower cartilage loss and improved joint mechanics in animal models.
- Reduced joint pain
- Improved mobility
- Cartilage protection (animal)
Researched for
- Knee osteoarthritis
- Shoulder and hip osteoarthritis
- Post-surgical joint recovery
- Tendinopathy (off-label)
Research notes
- Rabbit OA model (Kwon & Park 2015): AOD-9604 plus HA improved cartilage scores and function more than HA alone.
- HA viscosupplementation shows small-to-moderate pain benefit in knee OA meta-analyses; guideline support is mixed.
- No human trials of AOD-9604, alone or combined, for joint disease.
- Benefits reported by clinics are anecdotal and confounded by concurrent rehabilitation.
Reported side effects
- Post-injection joint pain or swelling
- Temporary effusion or warmth
- Rare joint infection
- Rare allergic reaction to HA
Contraindications & cautions
- Joint or skin infection at the site
- Active malignancy
- Bleeding disorder or anticoagulation (relative)
- Allergy to hyaluronic acid
- Pregnancy
Lab monitoring
- CBC and CMP
- ESR and CRP
- Joint imaging (X-ray or MRI)
- Pain/function scores (WOMAC or VAS)
